A useful tool for calculating the approximate time your PCR will take on any conventional thermocycler make or model. Use it to:

  • Determine how much time you can save by converting your existing PCR assay to a Fast PCR assay using the KAPA2G Fast PCR Kit
  • Optimize cycling parameters when using the KAPA2G Fast PCR Kit
  • Calculate how much time your PCR will take on different thermocyclers

Instructions for use:

  • Java version 1.6 or higher is needed to run the Calculator.
  • If your browser is set to disable ActiveX contents, allow the blocked content. The application takes a short while to load.
  • Fill out all the fields. Note the following:
    1. Use TAB or directional arrows to move between fields.
    2. All times are to be entered in seconds.
    3. Amplicon length is optional, except if you wish to use extension rate (sec/kb) instead of extension time.
    4. If you do not know the ramp rates of your thermocylcer (°C per second heating and cooling), a simple experiment may be set up to determine this (see below).
    5. The number of cycles is selected from a drop-down menu. Page Up and Page Down or arrows may also be used.
    6. All temperature fields must be filled.
    7. For a 2-step cycling profile, put the combined annealing/extension temperature in both the Annealing and Extension Temperature fields. Make the Annealing Time zero and enter the combined annealing/extension time in seconds in the Extension Time field.

To determine the ramp rates of your thermocycler:

Some thermocycler manufacturers average heating and cooling rates in their thermocycler specifications, but these are not always accurate. To determine the heating and cooling rates for your specific instrument, follow the 4 easy steps outline below:

  1. Program the thermocycler to cycle between 35°C and 95°C for 5 cycles.
  2. Using a stopwatch, measure the time in seconds it takes to heat from 35 °C to 95 °C in at least 3 of the cycles. Calculate the average time (in seconds).
  3. Divide 60°C by the average heating time to determine the average heating rate (°C per second).
  4. Repeat the process to determine the average cooling rate. This time, measure the time it takes to cool the block down from 95°C to 35°C in each cycle.
